Sample preparation for DNA extraction from bacteria
Use bacteria cells from overnight culture after centrifugation and discarding the media, or colonies from a culture plate. Use 107-109 cells or 5-20 mg of culture.

Sample preparation for extraction of DNA from plant (leaf, rice, potato)
Grind 5-20 mg leaves or rice or potato in a mortar and pestle in the presence of liquid nitrogen or after cooling it on dry ice. Use 5-20 mg of ground leaves for DNA isolation.
